Nucleotide sequence of the tissue-type plasminogen activator cDNA from human fetal lung cells.

چکیده

We have constructed igtiO cDNA libraries from the mRNA of the human fetal lung cell line. Flow 2000 (Japanese Cancer Research Resources Bank JCRB0067), and isolated two cDNA clones which together contain the complete coding region for a tissue-type plasminogen activator (t-PA). Sequence analysis reveals variant nucleotides compared with those reported for the cDNAs (1,2,3) and genoraic DNAs (4,5) derived frora other human cells. Our data agrees with that for the fetal liver genomic DNA determined by Degen et al. (5), except for two discrepancies (i.e., nudeotide positions 554 and 1694). In tha 3" untranslated region this sequence is different at 15 positions frora the Bowes melanomaderived cDNA sequence reported by Pennica et al. (1), whereas at only two positions frora that reported by Harris et al. (3). The initiation and termination codons are underlined.
